Living organs in electronic microsystems – Organ-on-Chip devices
We live in an era where the borders between, Pharma, MedTech and the Electronic Components and Systems industry (ECS) are fading. At the intersection between these domains new technologies are emerging that offer great opportunities for industry, as well as for society in the form of better and affordable healthcare. The pharmaceutical industry, for instance, has so far been primarily focussed on the development of chemical and biological compounds that are screened in-vitro in massive parallel biological assays, and in-vivo in hugely expensive clinical trials. Organ-on-chip devices combine human cells with microfluidic systems and sensors to form the basic elements of human organs. These devices offer better representative models for comprehension of disease mechanisms and drug development and can support the development of personal medicines.
Several biomedical research aim to analyse the behaviours of cell populations in artificial chemical environment, or precise monitoring their response for specific drug molecules. The microfluidic systems are applicable to reproduce the cellular micro-environment and to manipulate and characterise a limited but representative cell population. Our research aims to develop such application-specific microsystems which can support the high throughput and parallel sustenance, chemical treatment and physiological monitoring of artificial cell populations. Main challenge could be the elaboration, development, integration and validation of detection methods for cell responses. Our perspective is to develop microsystems to be able to accelerate the analysis of effects of drug agents – e.g. antibiotic resistance of bacteria.

Napjainkban a gyógyászat, a farmakológia és az elektronika határai egyre inkább elmosódnak. Ezen területek metszetében fejlődő innovatív technológiák hatalmas előrelépést jelentenek az ipar és a társadalom számára is a jobb és megfizethetőbb egészségügy irányába. A gyógyszeripar a kémiai és biológiai hatóanyagok kutatására, fejlesztésére és tesztelésére fókuszál, amelyek hatékonyságát párhuzamosítható in-vitro biológiai elemzésekkel, vagy roppant költséges in-vivo klinikai vizsgálatokkal validálják napjainkban. Ezek alternatívájaként jelentek meg az „Organ-on-Chip” eszközök, amelyek humán sejteket képesek mikrofluidikai rendszerekben kontroláltan életben tartani és érzékelőkkel folyamatosan figyelni, akár emberi szervek alapkultúráit megalkotva és a szerv alapvető funkcióját lemásolva. Ezek az eszközök jobb reprezentatív modellt adhatnak a betegségek mechanizmusainak megértéséhez, vagy gyógyszerhatóanyag tesztekhez, illetve a személyre szabott gyógyászat elősegítéséhez.
Számos orvosbiológiai kutatás célja sejtpopulációk mesterséges kémiai környezetben tapasztalható viselkedésének vizsgálatát, illetve sejtek adott hatóanyagra adott válaszának pontos követését. A mikrofluidikai rendszerek kifejezetten alkalmasak lehetnek a celluláris mikrokörnyezet valósághű reprodukálására, valamint a korlátozott, de reprezentatív méretű sejtpopulációk manipulálására, vizsgálatára. Kutatásunk célja olyan applikáció specifikus mikrorendszerek fejlesztése amelyek integrált mikrofluidikai és érzékelő rendszerek segítségével egyszerre oldják meg mesterséges sejtpopulációk, szövetek fenntartását, kémiai hatóanyaggal történő kezelését, valamint élettani viselkedésük folyamatos monitorozását. Különleges kihívást jelent a sejtválasznak megfelelő detektálási megoldások kidolgozása, megvalósítása és validálása. Távlati célunk olyan mikrorendszerek fejlesztése, amelyek méretüknél fogva felgyorsíthatják a gyógyszerhatóanyagok hatásainak elemzését – pl. antibiotikum rezisztencia kimutatását.
